Can Madmapper change the color of a fixture on tempo without changing the scene? In other words, without using auto-play.

If not, then can it turn the luminosity on/off on tempo?

Use MadMapper 4, you can create another cue bank and create a Cue Scheduler (just activating "Automate") that will trigger cues there on the beat on this bank specifically.
You can then still use the other Cue Bank for scenes.
However if you are using scenes, it will also contain the fixtures color (since scenes stores everything), you'd better switch to using cues in this case (right click a cue cell and choose "Add To Cue / All Objets". That's about the same as a scene then, except that you can remove fixture colors from those (in a single operation: select all your cues, type "color" in the search field, select the "Fixtures" node in the Cue Inspector and press delete)
